They certainly used to but I think the latest iTunes supports "reverse sync".

 

 

Found this manual method as well...

Put your iPod into Disk mode (hold Menu & Select for 5-6 secs, then when the Apple appears hold down select and Play/pause until the OK to disconnect and "tick" appears on the screen). Connect your iPod to your PC (it should then say do not disconnect). If iTunes starts, just get rid of it ASAP. Look at your iPod from My Computer. Make sure that you have enabled the option to view hidden files. In the root folder of the iPod there should be a hidden folder called iPod Control. Copy that entire folder to a convenient place on your PC (like your desktop) - this will take quite a while. Once the process has finished, disconnect your iPod (after doing the "safely remove hardware" bit).

 

Fire up iTunes, and get it to look for new music in the folder you just copied the stuff into. That should bring all of your music into iTunes. If you then want it to be arranged neatly on your PC, ask iTunes to do the "consolidate library" option from the Advanced menu. It will then copy all your music files into folders within the iTunes music folder, and arrange them by band, album etc.
